Erster Präsident der Bank war Oliver Wolcott (der von 1795-1800 Secretary of the Treasury war, also Finanzminister der USA). In den 1920er Jahren erfolgte der erste große Wachstums-Schub durch Fusion mit vielen anderen Instituten, so der Franklin Trust Co. und der Atlantic National Bank. Außerdem war die BoA Mitglied des Federal Reserve Systems und zur Verwahrung öffentlicher Gelder ermächtigt. 1928 übernahm der einflußreiche Bankier Amadeo Peter Giannini (1870-1949) aus San Francisco die Bank of Amerika und nutzte sie als Keimzelle für seine Expansionsziele.
